General description

DNA-dependent protein kinase catalytic subunit (EC 2.7.11.1; UniProt P78527; also known as DNA-PK catalytic subunit, DNA-PKcs, DNPK1, p460) is encoded by the PRKDC (also known as HYRC, HYRC1, IMD26, XRCC7) gene (Gene ID 5591) in human. DNA-PK is a heterotrimer of PRKDC and the Ku p70/YRCC6-p86/XRCC5 dimer. It acts as a molecular sensor for DNA damage and is involved in DNA non-homologous end joining required for double-strand break (DSB) repair and V(D)J recombination. Upon binding to DNA, it expresses its catalytic properties. DNA-PKcs falls into the phosphatidylinositol (PI) 3-kinase family, however, biochemical assays indicate that it phosphorylates proteins and has no detectable activity toward lipids. DNA-PKcs is displays high similarity to PI kinase family members involved in cell cycle control, DNA repair, and DNA damage responses. Deficiency of DNA-PKcs is linked to severe combined immunodeficiency characterized by reduced or absent T and B cells, recurrent candidiasis, and lower respiratory tract infections. Western Blotting Analysis: 0.5 µg/mL of this antibody detected DNA-PKcs in 10 µg of K562 cell lysate.

Specificity

Clone 3A9.1 detects DNA-PK catalytic subunit (DNA-PKcs), but not the regulatory subunit (Ku). Target epitope is present in both spliced isoforms of human DNA-PKcs reported by UniProt (P78527).

Stable for 1 year at 2-8°C from date of receipt.
